A brief inventory of sand flies (Diptera, Psychodidae) from the National Forest of the Rio Preto, state of the Espírito Santo, southeastern Brazil Biota Neotropica
Pinto,Israel de Souza; Tonini,João Filipe Riva; Ferreira,Adelson Luiz; Falqueto,Aloísio.
The inventories on sand flies are usually conducted for a long period in leishmaniasis-endemic regions. Thus, there is little data on the sand fly fauna in areas where no leishmaniasis cases have been reported. Therefore, we conducted an inventory to study the sand fly fauna in the National Forest of Rio Preto (NFRP). We carried out a brief inventory by using four CDC light traps in 2009 during winter and summer. We collected 1,403 sand flies belonging to 11 genera and 22 species. As expected, we did not find in the NFRP region sand flies species that have been reported as vectors of Leishmania. However, we found some species of sand flies with few records and that rarely have been reported in southeastern Brazil. Nevertheless, the diversity and species...

Actividad leishmanicida y antioxidante de extractos de Piper daniel-gonzalezii Trel. (Piperaceae) Plantas Medicinales
Cardona Galeano,Wilson; Robledo Restrepo,Sara M; Rojano,Benjamín Alberto; Alzate Guarin,Fernando; Muñoz Herrera,Diana Lorena; Saez Vega,Jairo.

Farahmand,Mahin; Nahrevanian,Hossein; Shirazi,Hasti Atashi; Naeimi,Sabah; Farzanehnejad,Zahra.
Cutaneous leishmaniasis (CL) is a widespread tropical infection which has a high incidence rate in Iran. Leishmania tropica, the causative agent of anthroponotic cutaneous leishmaniasis (ACL), and Leishmania major, which causes zoonotic cutaneous leishmaniasis (ZCL), are endemic in various parts of Iran with a high incidence rate. The aim of this study was to evaluate the reappraisal of the diagnosis and epidemiology of CL in Iran, by different clinical, parasitological and molecular assays among patients suspected of CL referred to the Department of Parasitology, at the Pasteur Institute of Iran during 2006-2009. Two hundred samples from patients with ulcerative skin lesions were collected, clinical analyses were applied, data questionnaire was completed...

Application of the mammalian glyceraldehyde-3-phosphate dehydrogenase gene for sample quality control in multiplex PCR for diagnosis of leishmaniasis J. Venom. Anim. Toxins incl. Trop. Dis.
Gonçalves,SC; Régis-da-Silva,CG; Brito,MEFC; Brandão-Filho,SP; Paiva-Cavalcanti,M.
Leishmaniasis is a neglected disease endemic in five continents. It is a severe disease that may lead to death, and its early detection is important to avoid severe damage to affected individuals. Molecular methods to detect Leishmania are considered alternatives to overcome the limitations presented by conventional methods. The aim of this study was to develop multiplex PCR systems able to detect small amounts of target DNA of Leishmania infantum and Leishmania braziliensis, and the gene coding for glyceraldehyde-3-phosphate dehydrogenase (G3PD) in mammals, enabling quality evaluation of the sample simultaneously with detection of the specific target. The systems created for G3PD recognition were combined with detection systems for L. infantum and L....

Cardiac and pulmonary alterations in symptomatic and asymptomatic dogs infectednaturally with Leishmania (Leishmania) chagasi BJMBR
Alves,G.B.B.; Pinho,F.A.; Silva,S.M.M.S.; Cruz,M.S.P.; Costa,F.A.L..
Fifteen symptomatic and seven asymptomatic dogs infected naturally with Leishmania chagasi were examined in order to identify the presence of parasites and changes in heart and lung. Histopathological, cytological, and immunohistochemical analyses were performed on samples of heart and lung tissues. An inflammatory reaction characterized by inflammatory mononuclear, perivascular and intermuscular infiltrates was observed in both symptomatic and asymptomatic animals on histopathological analysis of the heart. In the lung, there was thickening of the alveolar septa due to congestion, edema, inflammatory infiltrate, and fibroblast proliferation. A focal reaction was observed although a diffuse reaction was present in both groups. On cytological examination,...

Chalcones and N-acylhydrazones: direct analogues? Exploratory data analysis applied to potential novel antileishmanial agents BJPS
Rando,Daniela Gonçalves; Giarolla,Jeanine; Pasqualoto,Kerly Fernanda Mesquita; Ferreira,Elizabeth Igne.
Leishmaniasis is an important health and social problem for which there is limited effective therapy. Chalcones and N-acylhydrazones have been studied as promising antileishmanial agents in enzymatic inhibition and in vitro assays. Since these chemical classes of compounds also resemble each other structurally, it would be useful to investigate whether they share direct analogy. Exploratory data analysis was applied to a library of chalcones and nitrated N-acylhydrazones assayed against Leishmania donovani to investigate their similarity. Under the conditions applied in the present study, the two classes did not present functional or structural analogy.

Comparison of different primes for PCR-based diagnosis of cutaneous leishmaniasis BJID
Oliveira,Diego Molina de; Lonardoni,Maria Valdrinez Campana; Teodoro,Ueslei; Silveira,Thais Gomes Verzignassi.
OBJECTIVE: The objective of this study was to analyze different primers that are commonly used in epidemiological studies for the detection of Leishmania DNA by PCR, and to compare them to the conventional direct parasite search for American cutaneous leishmaniasis (ACL) diagnosis. MATERIAL AND METHODS: Five pairs of primers, four of them derived from Leishmania kDNA sequences (MP3H-MP1L; B1-B2; LBF1-LBR1; 13A-13B), and one derived from the SL RNA (mini-exon) gene repeat (LU5A-LB3C), reported previously, were used. RESULTS: The MP3H-MP1L primers were the best at amplifying the DNA, detecting 2 fg of Leishmania spp. DNA. The 13A-13B primers presented the worst performance, detecting 512 x 10³ fg of DNA. CONCLUSION: The wide variation in the analytical...

Comparison of the specificity of PCR and the histopathological detection of leishmania for the diagnosis of American cutaneous leishmaniasis BJMBR
Medeiros,A.C.R.; Rodrigues,S.S.; Roselino,A.M.F..
More precise and rapid diagnostic methods for American cutaneous leishmaniasis (ACL) are necessary because of the growing number of cases observed in Brazil, including the northeastern region of the State of São Paulo. We applied PCR to 54 skin or mucosal biopsies from patients with a clinical and/or laboratory diagnosis of ACL using primers 13A and 13B, with positive results being obtained for 82% of the samples. When the PCR results were compared to those of histopathological leishmania detection, PCR showed superior results with 81.5% sensitivity and 95% CI of 68.0-95.1%. The Montenegro skin test (MST) was positive in 88.7% of patients. Since MST cannot be used as a diagnostic tool in endemic areas, the present results strongly suggest the use of PCR...

Cytokine profile and pathology in human leishmaniasis BJMBR
Ribeiro-de-Jesus,A.; Almeida,R.P.; Lessa,H.; Bacellar,O.; Carvalho,E.M..
The clinical spectrum of leishmaniasis and control of the infection are influenced by the parasite-host relationship. The role of cellular immune responses of the Th1 type in the protection against disease in experimental and human leishmaniasis is well established. In humans, production of IFN-g is associated with the control of infection in children infected by Leishmania chagasi. In visceral leishmaniasis, an impairment in IFN-g production and high IL-4 and IL-10 levels (Th2 cytokines) are observed in antigen-stimulated peripheral blood mononuclear cells (PBMC). Moreover, IL-12 restores IFN-g production and enhances the cytotoxic...

Distribution of liposome-encapsulated antimony in dogs BJMBR
Schettini,D.A.; Costa Val,A.P.; Souza,L.F.; Demicheli,C.; Rocha,O.G.F.; Melo,M.N.; Michalick,M.S.M.; Frézard,F..
The achievement of complete cure in dogs with visceral leishmaniasis is currently a great challenge, since dogs are the main reservoir for the transmission of visceral leishmaniasis to humans and they respond poorly to conventional treatment with pentavalent antimonials. In order to improve the efficacy of treatment, we developed a novel formulation for meglumine antimoniate based on the encapsulation of this drug in freeze-dried liposomes (LMA). The aim of the present study was to evaluate the biodistribution of antimony (Sb) in dogs following a single intravenous bolus injection of LMA. Four healthy male mongrel dogs received LMA at 3.8 mg Sb/kg body weight and were sacrificed 3, 48 and 96 h and 7 days later. Antimony was determined in the blood, liver,...

Distribution of the assemblage of phlebotomine sandflies (Diptera: Psychodidae) along an environmental gradient Biota Neotropica
Antonini,Yasmine; Fernandes,G. Wilson; Barata,Ricardo Andrade.
Abstract The geographical distribution of leishmaniasis has undoubtedly expanded, and is now being reported to be in areas that were previously non-endemic. This study therefore aimed to investigate whether there is any variation in the structure and in the composition of phlebotomine communities along an environmental gradient - from a mesic to a xeric environment, across different climatic periods (rainy and dry seasons). Furthermore, we test whether there were differences in sex ratio of sand flies between wet and dry environments. A total of 535 specimens of sand flies belonging to 18 species were recorded. Richness, abundance, and composition of species were significatively higher in the xeric environment (semidecidual Forest) compared to the mesic...

Estudos Sobre Leishmaniose: I. Primeiros Casos de Leishmaniose Espontânea Observados em Cobáios BABT
Medina,Heitor.
The author describes the first cases of spontaneous leishmaniasis in guinea-pigs, as verified for the first time, in Curitiba, at the Institute of Biology and Technological Research. Studying the inoculability of Paracoccidioide brasiliensis in guinea-pigs, he found animals which presented tumoral lesions in the ears and peritesticular skin. The examination of these tumors, mixed with a small drop of Giemsa's original solution, plus the exam of the rubbings of the same material, coloured by the May-Grunwald Giemsa after fixation by Heidenhein's solution and the histological exams of the cuts in the tumoral lesions revealed the presence of elements which possessed the same morphology and aspect of the LEISHMAN-DONOVANI corpuscles. In the meanwhile, a...
